Does it have any significant effect if a planet falls in a sign, but it's in the opposite sign's house? Or even if the sign the planet is in is opposite that planet's ruler?
Yes. If there is an "opposite" situation in the chart, then the planet or sign is in the "opposite" place from where it would "like" to be. This is called being in "detriment" and indicates that there is a "challenge" to the planet or sign. A planet is in "detriment" when it is in the house opposite to the one it "fits" best with and a sign is in "detriment" when it is modifies the planet opposite the sign it naturally rules.
Let's look at this using "key words" and your examples to make things easier (if you have questions about this, look at Step 5 at http://www.geocities.com/wilsontctc to follow along):
Sun (self-expression) focused in the 11th house (friends)
The house of "self-expression" is opposite the 11th house. So Sun is in the "opposite" house", which means Sun is "challenged" in its energy (i.e., "in detriment").
Mercury (daily work) in the 12th house (spirituality)
The house of "daily work" is opposite the 12th house. So Mercury is in the "opposite" house", which means Mercury is "challenged" in its energy (i.e., "in detriment")
These are all examples of planets in detriment.
Capricorn (duty) modifying Uranus (friends)
The keyword opposite the "friends" keyword is "self-expression" (as mentioned above, so this is NOT an example of a sign in detriment.
Saturn is the traditional ruler of Aquarius. So, if we use Saturn as the ruler of Aquarius, Aquarius modifying Saturn is Saturn in the sign it rules. This also is NOT an example of a sign in its detriment.
If you take a look over Step 5, and take a look at which signs, planets, and houses are opposite each other, this will make understading detriments MUCH easier.
Easily,
